ESMFold2 design

ESMFold2 design is a no-install online antibody-fragment and minibinder design tool you can run through tools.ranomics.com on a dedicated GPU. All six binding loops of the scFv are designed together in one pass against your target rather than one at a time, and the same run can make small de novo binders instead.

Design inputs
The first seed in the sweep. Different seeds yield different designs.
Parallel sweep size (1 to 64). All seeds run on their own H100 in parallel, so a 16-seed sweep finishes in the same wall-clock as one seed. Cost scales linearly.
Designs per seed (1 to 6). All share one H100 pass per seed, so a higher batch multiplies candidates at the same wall-clock. Default 3 balances yield against the drop rate from the iPTM / pI gates.

ESMFold2 design backpropagates through the structure predictor to jointly optimize sequence and binding pose. Strict-pass designs (iptm > 0.75 and pI < 6) are the ones worth ordering. The scaling critics were not used — only the real iPTM is calibrated; the proxy is informative only.
